TOX4
TOX4 (TOX High Mobility Group Box Family Member 4) is a protein that in humans is encoded by the TOX4 gene. This protein is part of the TOX family, which is characterized by containing a high mobility group (HMG) box domain. The TOX proteins play critical roles in the regulation of DNA binding and thus are involved in the control of gene expression, particularly in the immune system and during cell differentiation and development.
Function[edit]
The TOX4 protein is involved in various cellular processes, including the regulation of gene expression, cell cycle progression, and the maintenance of chromatin structure. It is known to interact with the PARP1 (Poly [ADP-ribose] polymerase 1) protein, playing a role in the repair of DNA damage and maintaining genomic stability. Through its interaction with PARP1, TOX4 participates in the regulation of transcription by modulating the structure of chromatin, thereby influencing the accessibility of genes to the transcriptional machinery.
Clinical Significance[edit]
While the full clinical significance of TOX4 is still under investigation, alterations in the expression or function of TOX4 have been implicated in various cancers. Its role in DNA repair and gene expression regulation suggests that TOX4 could be involved in the development and progression of cancer through mechanisms such as the promotion of genomic instability or the dysregulation of genes critical for cell growth and survival. Further research is necessary to fully understand the potential of TOX4 as a biomarker for cancer or as a target for therapeutic intervention.

The TOX4 gene is located on chromosome 14q32.33 in humans. It is expressed in various tissues, with significant levels observed in the testis, brain, and heart. The expression of TOX4, like other members of the TOX family, is tightly regulated, suggesting its importance in the maintenance of normal cellular functions and its potential involvement in disease when dysregulated.
