Amtolmetin Guacil

Chemical structure of Amtolmetin Guacil

Amtolmetin guacil is a non-steroidal anti-inflammatory drug (NSAID) used primarily for its analgesic and anti-inflammatory properties. It is a prodrug of tolmetin, designed to reduce gastrointestinal side effects commonly associated with NSAIDs.

Pharmacology

Amtolmetin guacil is metabolized in the body to its active form, tolmetin, which inhibits the cyclooxygenase (COX) enzymes, particularly COX-1 and COX-2. This inhibition leads to a decrease in the synthesis of prostaglandins, which are mediators of inflammation and pain.

Mechanism of Action

The primary mechanism of action of amtolmetin guacil involves the inhibition of the COX enzymes. By reducing prostaglandin synthesis, it alleviates symptoms of pain and inflammation. The prodrug form is intended to minimize direct irritation to the gastrointestinal tract, a common side effect of NSAIDs.

Clinical Uses

Amtolmetin guacil is used in the management of various conditions characterized by pain and inflammation, such as osteoarthritis, rheumatoid arthritis, and other musculoskeletal disorders. It is also used for the relief of mild to moderate pain.

Side Effects

Common side effects of amtolmetin guacil include nausea, dyspepsia, and headache. As with other NSAIDs, there is a risk of gastrointestinal bleeding, ulceration, and renal impairment.

Contraindications

Amtolmetin guacil is contraindicated in patients with known hypersensitivity to tolmetin or other NSAIDs, and in those with a history of asthma, urticaria, or other allergic-type reactions after taking aspirin or other NSAIDs.
