CRTC1
CRTC1 (CREB-regulated transcription coactivator 1) is a protein that in humans is encoded by the CRTC1 gene. It is a member of the CREB (cAMP response element-binding protein) regulated transcription coactivator family. This protein has been shown to interact with and augment the activity of adenylate cyclase and cAMP/protein kinase A pathway that mediates signal-dependent activation of CREB.
Function[edit]
CRTC1 is a transcriptional coactivator that regulates the genes involved in energy homeostasis, somatic growth, and adipogenesis. It is also involved in the regulation of long-term memory and synaptic plasticity in the hippocampus. CRTC1 is phosphorylated by AMP-activated protein kinase (AMPK) and is thus regulated by cellular energy status.
Clinical significance[edit]
Mutations in the CRTC1 gene have been associated with Sotos syndrome, a genetic condition causing physical overgrowth during the first years of life. CRTC1 has also been implicated in the development of cancer, particularly lung cancer and endometrial cancer.
