Duodenal cytochrome B

Duodenal cytochrome b (Dcytb) is a protein that in humans is encoded by the CYBRD1 gene. This protein is a member of the cytochrome b family and is primarily located in the duodenum, the first section of the small intestine. Dcytb plays a crucial role in iron absorption by reducing ferric iron (Fe^3+) to ferrous iron (Fe^2+), which is a more soluble form that can be easily absorbed by the intestinal lining.

Function

Duodenal cytochrome b is an integral membrane enzyme found on the brush border of the duodenum. It functions as a ferric reductase, converting dietary iron in the form of insoluble ferric iron into the soluble ferrous form. This process is essential for the maintenance of normal iron homeostasis in the body, as it facilitates the absorption of iron, which is a critical component of hemoglobin and various other enzymes involved in electron transport and DNA synthesis.

Genetic Expression

The CYBRD1 gene, located on chromosome 2, encodes the Dcytb protein. Expression of this gene is upregulated in response to iron deficiency, highlighting its role in iron metabolism. Regulatory mechanisms involve the hypoxia-inducible factors (HIFs) pathway, which adjusts gene expression in response to iron availability and oxygen levels.
