Lymphangiogenesis

Lymphangiogenesis (pronunciation: lim-fan-gee-oh-gen-esis) is the process of lymphatic vessel formation from pre-existing lymphatic vessels. This process is crucial for the development of the lymphatic system and plays a significant role in various pathological conditions, including cancer and inflammation.

Etymology

The term 'Lymphangiogenesis' is derived from three Greek words: 'lymph' meaning water, 'angeion' meaning vessel, and 'genesis' meaning origin or creation. Thus, it refers to the creation or formation of lymphatic vessels.

Process

Lymphangiogenesis begins with the differentiation of lymphatic endothelial cells (LECs) from venous endothelial cells (VECs). This process is regulated by several growth factors, including vascular endothelial growth factor C (VEGF-C) and vascular endothelial growth factor D (VEGF-D). These growth factors bind to the VEGF receptor 3 (VEGFR-3) on the surface of LECs, triggering a series of intracellular events that lead to the proliferation, migration, and tube formation of LECs.

Role in Disease

Abnormal lymphangiogenesis is associated with a variety of diseases. In cancer, for example, tumor cells can stimulate lymphangiogenesis to facilitate their spread to lymph nodes and distant organs, a process known as lymphatic metastasis. In inflammatory diseases, increased lymphangiogenesis can contribute to the resolution of inflammation by enhancing the clearance of inflammatory cells and mediators.
