LAMP3
LAMP3 (Lysosomal Associated Membrane Protein 3), also known as DC-LAMP or CD208, is a protein that in humans is encoded by the LAMP3 gene. It is a member of the lysosome associated membrane protein (LAMP) family, which is characterized by a highly glycosylated luminal domain and a relatively short cytoplasmic tail.
Function[edit]
LAMP3 is primarily expressed in dendritic cells, where it is involved in the formation and maintenance of the endosomal compartment. It is also involved in the process of antigen presentation, which is crucial for the activation of the immune response. LAMP3 is also found in other cell types, including cancer cells, where it may play a role in tumor progression and metastasis.
Structure[edit]
The LAMP3 protein is a type I transmembrane protein, meaning it has a single transmembrane domain with its N-terminus on the outside of the cell and its C-terminus on the inside. The luminal domain of LAMP3 is heavily glycosylated, which is thought to protect the protein from degradation in the harsh environment of the lysosome.
Clinical significance[edit]
Alterations in the expression of LAMP3 have been associated with various diseases, including cancer, autoimmune diseases, and infectious diseases. In cancer, overexpression of LAMP3 has been linked to increased tumor aggressiveness and poor prognosis. In autoimmune diseases, LAMP3 may play a role in the presentation of self-antigens, leading to an inappropriate immune response. In infectious diseases, LAMP3 may be involved in the presentation of pathogen-derived antigens, leading to an immune response against the pathogen.
