GLUT8
GLUT8 is a member of the glucose transporter family, which are integral membrane proteins that facilitate the transport of glucose across the plasma membrane. GLUT8 is encoded by the SLC2A8 gene and is part of the class III facilitative glucose transporters.
Structure
GLUT8, like other glucose transporters, is characterized by 12 transmembrane helices. These helices form a channel through which glucose can be transported. The structure of GLUT8 allows it to specifically bind and transport glucose molecules, although its exact substrate specificity and transport kinetics can vary.
Expression
GLUT8 is expressed in a variety of tissues, including the brain, testis, and placenta. Its expression in the brain suggests a role in neuronal glucose uptake, while its presence in the testis and placenta indicates a potential role in reproductive physiology.
Function
The primary function of GLUT8 is to facilitate the transport of glucose across cell membranes. Unlike some other glucose transporters, GLUT8 is thought to be involved in the transport of glucose into intracellular compartments, such as the endoplasmic reticulum. This intracellular transport is crucial for various cellular processes, including glycosylation and energy metabolism.
Regulation
GLUT8 activity is regulated by several factors, including insulin and AMP-activated protein kinase (AMPK). Insulin can stimulate the translocation of GLUT8 to the plasma membrane, enhancing glucose uptake. AMPK, a key energy sensor in cells, can also modulate GLUT8 activity in response to changes in cellular energy status.
Clinical Significance
Alterations in GLUT8 expression or function have been implicated in various diseases. For example, dysregulation of GLUT8 may contribute to insulin resistance and type 2 diabetes. Additionally, GLUT8 has been studied in the context of cancer, where altered glucose metabolism is a hallmark of tumor cells.
