AS03

AS03 is an adjuvant used in vaccines to enhance the body's immune response to an antigen. It is a squalene-based immunologic adjuvant that is used in various vaccines, including those for influenza and hepatitis.

Composition

AS03 contains squalene, DL-α-tocopherol, and polysorbate 80. Squalene is a naturally occurring oil that is present in humans, animals, and plants. DL-α-tocopherol, also known as vitamin E, is an antioxidant that protects cells from damage. Polysorbate 80 is a surfactant and emulsifier used in foods, cosmetics, and pharmaceuticals.

Mechanism of Action

The exact mechanism of action of AS03 is not fully understood. However, it is believed to work by stimulating the body's immune system to produce a stronger and longer-lasting response to the antigen in the vaccine. This is achieved by enhancing the presentation of the antigen to the immune cells, thereby increasing the production of antibodies and T cells.

Use in Vaccines

AS03 has been used in several vaccines, including the Pandemrix vaccine for H1N1 influenza and the Fendrix vaccine for hepatitis B. It is also being studied for use in vaccines for other diseases, such as malaria and HIV.

Safety and Side Effects

AS03 has been shown to be safe and well-tolerated in clinical trials. The most common side effects are mild and temporary, such as pain and swelling at the injection site, fatigue, headache, and muscle pain. However, like all medicines, vaccines with AS03 can cause side effects, although not everybody gets them.
