The medical school was initially established as the Durham University College of Medicine in 1834. It was originally located in the Barber Surgeons' Hall on Houston Street, which played a significant role in the early development of medical education in Newcastle.

In 1851, the medical school became part of the University of Durham, and in 1937, it was incorporated into King's College, Durham. The school continued to grow and develop, eventually becoming part of the newly established Newcastle University in 1963.
