TRAF6

TRAF6 or TNF receptor associated factor 6 is a protein that in humans is encoded by the TRAF6 gene. It is a member of the TNF receptor associated factor (TRAF) protein family. TRAF proteins associate with, and mediate the signal transduction from, members of the TNF receptor (TNFR) superfamily. This protein directly interacts with TNFR superfamily members and participates in their signal transduction.

Function

TRAF6 is a signal transducer in the NF-kappaB pathway that activates IKK (IκB kinase). It is involved in the stimulation of NF-kappaB and JNK (c-Jun N-terminal kinase). TRAF6 also regulates the differentiation and function of osteoclasts and lymphocytes, and the activation of immune responses.

Clinical significance

Mutations in the TRAF6 gene are associated with various diseases, including Paget's disease of bone and frontotemporal dementia. TRAF6 has also been implicated in the pathogenesis of cancer, autoimmune diseases, and infectious diseases.

Interactions

TRAF6 has been shown to interact with a number of proteins, including IRAK1, IRAK4, TAK1, and TAB2. These interactions are crucial for the activation of downstream signaling pathways.
