FZD2 or Frizzled Class Receptor 2 is a protein that in humans is encoded by the FZD2 gene. This gene is a member of the frizzled gene family, which is involved in the Wnt signaling pathway. The Wnt signaling pathway plays a critical role in embryonic development, cell proliferation, migration, and differentiation. Dysregulation of this pathway has been implicated in a variety of diseases, including cancer.

Function[edit]

FZD2 is a G protein-coupled receptor that interacts with Wnt proteins to initiate the Wnt signaling pathway. Upon binding to a Wnt protein, FZD2 activates the Dishevelled (DVL) proteins inside the cell, which then inhibits the degradation of beta-catenin. Stabilized beta-catenin can enter the nucleus and regulate the expression of target genes that are involved in cell fate determination, proliferation, and survival.

Clinical Significance[edit]

Alterations in the expression or function of FZD2 have been observed in various types of cancer, including colorectal, breast, and gastric cancers. These alterations may lead to aberrant activation of the Wnt signaling pathway, promoting tumor growth and metastasis. Therefore, FZD2 is considered a potential target for cancer therapy.

In addition to its role in cancer, FZD2 has been implicated in other pathological conditions such as fibrosis and osteoarthritis. Research is ongoing to fully understand the role of FZD2 in these diseases and to develop targeted therapies.

Genetics[edit]

The FZD2 gene is located on chromosome 7q21.1 in humans. It consists of several exons that encode the FZD2 protein. Genetic variations in the FZD2 gene may influence the risk of developing certain diseases, although more research is needed to clarify these associations.
