TNFSF12 (Tumor Necrosis Factor Superfamily Member 12), also known as TWEAK (TNF-related weak inducer of apoptosis), is a protein that in humans is encoded by the TNFSF12 gene. It is a member of the tumor necrosis factor (TNF) ligand family and is a ligand for the FN14/TWEAKR receptor.

Function

TNFSF12 is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This protein is a ligand for the FN14/TWEAKR receptor. This cytokine has overlapping signaling functions with other members of the TNF superfamily. It can induce apoptosis via multiple pathways of cell death in a cell type-specific manner. This cytokine is also found to promote proliferation and migration of endothelial cells, and thus acts as a regulator of angiogenesis.

Clinical Significance

Aberrant expression of TNFSF12 has been noted in various diseases, including cancer, autoimmune diseases, and inflammatory diseases. It has been suggested as a potential therapeutic target in these conditions.

Interactions

TNFSF12 has been shown to interact with TNFRSF12A.
