PK/PD Models

PK/PD models (pronounced as P-K/P-D models), also known as Pharmacokinetic/Pharmacodynamic models, are mathematical models used in pharmacology to predict the effect of a drug on a biological system.

Pronunciation

  • PK: /piːˈkeɪ/
  • PD: /piːˈdiː/

Etymology

The term PK/PD models is derived from the words Pharmacokinetics (PK) and Pharmacodynamics (PD). Pharmacokinetics, from the Greek words pharmakon (drug) and kinesis (movement), is the study of how a drug is absorbed, distributed, metabolized, and excreted by the body. Pharmacodynamics, from the Greek words pharmakon (drug) and dynamis (power), is the study of the biochemical and physiological effects of drugs and their mechanisms of action.

Definition

PK/PD models are used to describe the time course of effect intensity in response to administration of a drug. The PK part of the model defines the relationship between the drug dose and the drug concentration in the body over time. The PD part of the model describes the relationship between the drug concentration and the effect of the drug.

Related Terms

  • Pharmacokinetics: The study of how a drug is absorbed, distributed, metabolized, and excreted by the body.
  • Pharmacodynamics: The study of the biochemical and physiological effects of drugs and their mechanisms of action.
  • Dose-response relationship: The relationship between the dose of a drug and the response produced.
  • Drug metabolism: The process by which the body breaks down and converts medication into active chemical substances.
  • Bioavailability: The proportion of a drug or other substance which enters the circulation when introduced into the body and so is able to have an active effect.
