Nisterime' is a synthetic compound classified as an anabolic-androgenic steroid (AAS), known by its International Nonproprietary Name (INN) as Nisterime. It is a derivative of dihydrotestosterone (DHT), a potent natural androgen, and is chemically identified as 2α-chloro-4,5α-dihydrotestosterone 3-O-(p-nitrophenyl)oxime or 2α-chloro-5α-androstan-17β-ol-3-one 3-O-(p-nitrophenyl)oxime. Despite its development, Nisterime was never introduced to the market for clinical use.

Chemical Structure and Derivatives

Nisterime's structure incorporates a 2α-chloro modification and a 3-O-(p-nitrophenyl)oxime group to the dihydrotestosterone backbone. This alteration aims to enhance its anabolic-androgenic properties. Additionally, a derivative of Nisterime, known as Nisterime Acetate (developmental code name ORF-9326), exists as the C17α acetate ester form. Nisterime Acetate was explored as a postcoital contraceptive but, similar to Nisterime, was never marketed.

Mechanism of Action

As an AAS, Nisterime's mechanism of action involves modulating the androgen receptor, similar to other steroids in its class. This interaction can promote protein synthesis, muscle growth, and other androgenic effects. However, specific details about Nisterime's activity, selectivity, and potency compared to DHT and other AAS remain less documented due to its lack of commercial development.

Clinical Development and Discontinuation

Both Nisterime and its acetate ester were subjects of research for potential therapeutic applications. Nisterime Acetate's investigation as a postcoital contraceptive highlights the exploration of AAS derivatives for reproductive health solutions. The discontinuation of these compounds' development and their absence from the market could be attributed to various factors, including efficacy concerns, safety profiles, or the emergence of more favorable alternatives.
